    Ministerial influence on the machinery of government: insights on the Inside

    The structure and organisation of the machinery of government is key to theambitions of political coalitions. When portfolio allocation and agencificationare a function of political choice, political volatility should also affect internalstructure of government administrations. This study tests the effects of politicalturnover of individual ministers and of the political ideology of coalitionson a dataset of intra-ministerial changes in Dutch ministries between 1980and 2014. Findings indicate that the turnover of political heads of departmentsand the shifts in policy preferences between successive coalitionsindeed affects the internal structure of ministerial departments. Political variableshave a strong impact, particularly changes in the left–right position ofthe government. A clear pattern for how precisely politics affect the structuraldesign of public organisations remains absent, in spite of the robustness ofthe findings. Most ministries experience significant effect of executive turnover,sometimes increasing the hazards of intra-organisational transitions andsometimes increasing stability. It turns out that ministers can substantially rearrangetheir organisations in line with their policy preferences but do notnecessarily do so.     NikR mediates nickel-responsive transcriptional induction of urease expression in Helicobacter pylori

    The important human pathogen Helicobacter pylori requires the abundant expression and activity of its urease enzyme for colonization of the gastric mucosa. The transcription, expression, and activity of H. pylori urease were previously demonstrated to be induced by nickel supplementation of growth media. Here it is demonstrated that the HP1338 protein, an ortholog of the Escherichia coli nickel regulatory protein NikR, mediates nickel-responsive induction of urease expression in H. pylori. Mutation of the HP1338 gene (nikR) of H. pylori strain 26695 resulted in significant growth inhibition of the nikR mutant in the presence of supplementation with NiCl(2) at > or =100 microM, whereas the wild-type strain tolerated more than 10-fold-higher levels of NiCl(2). Mutation of nikR did not affect urease subunit expression or urease enzyme activity in unsupplemented growth media. However, the nickel-induced increase in urease subunit expression and urease enzyme activity observed in wild-type H. pylori was absent in the H. pylori nikR mutant. A similar lack of nickel responsiveness was observed upon removal of a 19-bp palindromic sequence in the ureA promoter, as demonstrated by using a genomic ureA::lacZ reporter gene fusion. In conclusion, the H. pylori NikR protein and a 19-bp operator sequence in the ureA promoter are both essential for nickel-responsive induction of urease expression in H. pylori

    Identifier et expliquer les changements dans la structure des bureaucraties de l’État central et les déterminants de la survie des organisations publiques individuelles sont deux domaines de recherche étroitement liés dans l’administration publique. Nous cherchons à combler le fossé entre ces deux principaux courants d’études du changement organisationnel en présentant une nouvelle approche de la collecte de données sur l’historique des événements pour les organisations publiques. Nous avons développé ce cadre dans le cadre du projet « Structure et organisation des gouvernements », qui vise à cartographier l’ensemble des bureaucraties de l’État central dans trois pays d’Europe occidentale. Notre approche est suffisamment flexible pour décrire les macro-tendances des populations d’organisations du secteur public et pour expliquer ces tendances en analysant l’histoire des événements des organisations qu’elles comprennent. En plus de présenter notre cadre et la manière dont nous l’avons appliqué pour créer cet ensemble de données, nous présentons également quelques premières comparaisons transnationales de la répartition des types d’événements enregistrés, et soulignons les premiers résultats et les pistes prometteuses pour des recherches ultérieures.NWOORA 464-13-113The politics and administration of institutional changeSecurity and Global Affair

    Long-term outcome after the arterial switch operation: 43 years of experience

    OBJECTIVES: The objective of this study was to assess our 43-year experience with arterial switch operation (ASO) for transposition of the great arteries (TGA) by analysing cardiac outcome measures (hospital and late mortality, reoperations and catheter interventions, significant coronary artery obstruction) and to identify risk factors for reoperation and catheter interventions.METHODS: A total of 490 patients who underwent ASO for TGA from 1977 to 2020 were included in this retrospective, single-centre study. Data on reoperation and catheter intervention of hospital survivors were estimated by the Kaplan-Meier method and compared using a long-rank test. Risk factors for reoperation and/or catheter intervention were assessed by multivariate Cox regression analysis.RESULTS: Hospital mortality occurred in 43 patients (8.8%), late death in 12 patients (2.9%) and 43 patients were lost to follow-up. Median follow-up time of 413 hospital survivors was 15.6 (interquartile range 7.0-22.4) years. Reoperations were performed in 83 patients (117 reoperations). Neoaortic valve regurgitation with root dilatation was the second most common indication for reoperation (15/83 patients, 18.1%) after right ventricular outflow tract obstruction (50/83 patients, 60.2%). Risk factors for any reoperation on multivariable analysis were: TGA morphological subtype [TGA with ventricular septal defect: hazard ratio (HR)=1.99, 95% confidence interval (CI) 1.18-3.36; P=0.010 and Taussig-Bing: HR=2.17, 95% CI 1.02-4.64; P=0.045], aortic arch repair associated with ASO (HR=3.03, 95% CI 1.62-5.69; P=0.001) and a non-usual coronary artery anatomy (HR=2.41, 95% CI 1.45-4.00; P=0.001). One hundred and one catheter interventions were performed in 54 patients, usually for relief of supravalvular pulmonary stenosis (44/54 patients, 81.5%) or arch obstruction (10/54 patients, 18.5%). Main risk factor for catheter intervention on multivariable analysis was aortic arch repair associated with ASO (HR=2.95, 95% CI 1.37-6.36; P=0.006). Significant coronary artery stenosis was relatively uncommon (9/413 patients, 2.2%) but may be underrepresented.CONCLUSIONS: Patients after ASO typically have good long-term clinical outcomes but reoperations and interventions remain necessary in some patients. Neoaortic valve regurgitation with root dilatation is the second most common indication for reoperation after right ventricular outflow tract obstruction and an increasing need for neoaortic valve and root redo surgery in future is to be expected.Cardiolog

    Spatial representation for navigation in animats

    This article considers the problem of spatial representation for animat navigation systems. It is proposed that the global navigation task, or "wayfinding, " is best supported by multiple interacting subsystems, each of which builds its own partial representation of relevant world knowledge. Evidence from the study of animal navigation is reviewed to demonstrate that similar principles underlie the wayfinding behavior of animals, including humans. A simulated wayfinding system is described that embodies and illustrates several of the themes identified with animat navigation. This system constructs a network of partial models of the quantitative spatial relations between groups of salient landmarks. Navigation tasks are solved by propagating egocentric view information through this network, using a simple but effective heuristic to arbitrate between multiple solutions
